Agomelatine is a new type of antidepressant. Its mechanism of action breaks through the traditional monoamine transmitter system. It activates melatonin receptors, MT1 and MT2, and antagonizes 5-HT2C receptors. It can improve sleep quality and restore biological rhythm while having antidepressant effect; among them, by antagonizing the 5-HT2C receptor of the postsynaptic membrane, it can increase the release of DA and NE in the prefrontal cortex and play an antidepressant role. When MT agonism and 5-HT2C receptor antagonism exist at the same time, a unique synergistic effect can be produced to promote the release of more DA and NE in the PFC brain area, further strengthening the antidepressant effect. In addition, agomelatine can also promote the release of brain-derived neurotrophic factor in the PFC and block the stress-induced release of glutamate in the amygdala brain region.
